This game is much more complex and in depth. Every role has multiple powers, and they interact with each other very differently.
People also take this game more seriously than TOS.

This also has more abilities per character, for example knight can guard someone ( like tos body guard) OR attack someone ( like tos vigilante)
The cult/unseen faction are both like vampires, you can convert people ( and also kill them) but when you convert people they still have abilities ( such as killing things / occupying, etc depending on their original class)
The King is kind of like the mayor ( everyone knows who the king is) votes count as extra etc, but he can check on peoples faction at night, or send guards to protect people, and himself. The dynamic here is really what is different because sometimes the king is evil, good, or neutral. So that dynamic is a big difference, sometimes the Blue Dragon ( the town) will have to execute the king if they suspect he is evil. And when the king is dead a good, neutral, or evil player can step up to the throne.

A lof of the classes are the converted counterparts, the converted ones are usually the same with a few exceptions ( I think converted hunter has some unique abilities)
So when you are the mafia/vampires ( cult, or unseen) choosing who you want to convert, or kill is really strategic.
Most characters have multiple abilities ( except for prince which is pretty much exactly like jailor, but with only 3 executions) and maybe a few passive abilties.
Edit: http://tol.wikia.com/wiki/The_Herbalist look at converted physician, he has some epic abilties, fake healing someone, and changing the class of someone who dies to cause confusion. So you can coordinate with assassin and change that targets class.
Also on this if you die early on there is a good chance the physician will revive you as a death knight, and you can guard, or kill other players for a few nights ^-^.
