Report a translation problem
After you set the game to 1 cell difficulty through this, beating the hand of the king will drop your second cell, then when you defeat it on 2 cells you get your third and so on.
EDIT: welp, as i was expecting im just retarded, i kept activating the tube thing and see that there was nothing in there and was just closing it.
I didnt know i had to press right to see my boss steam cell T_T
If you look at the sides of the tube you will see keys or buttons to change how many cells you want in the tube (left and right d-pad for gamepads, A and D for keyboard by default).
I just stumbled upon trying to "Restart" in the pause menu. After restarting, suddenly there was a prompt available at the glass tube and I was able to select the harder difficulty at that point.
Some extra dialogue with the tutorial girl starting new game would be enough I guess... Instead you have basic controls to learn.
The weirdest thing about the tube option and its overall lack of intuitiveness is that the arrows to switch difficulty are easier to see on keyboard... like, the hint that you can change that screen is EASIER to see on a "non-recommended" playstyle.
As it stands, the tube is there, you can interact with it, and it says "No boss cells."
Change that to 0 (zero) boss cells, and put some pictures of arrows on the side of the menu. Easy.
