Cooldown (grenades or other powers) can have a significant impact if you have two powers affected by it, and there are plenty of other really good mutations to unlock.
But unless you are either really good and/or in a high difficulty (3+ boss cells for example) the first pick will usually go to "Ygdar Orus Li Ox", the mutation that revives you on your first death of the run because of how potent it is (plus you can't pick it if you don't take it as your first).
Personally, I tend to go for either grenade or powers cooldown reduction (depending on my main stat/gear), or parting gift/sadism if my findings in the first level makes a good use of those.
And overall, I rarely ever pick combo even later on unless I get a good combination of gear that favours speed (swift sword and a bonus that gives the swiftness after a single kill), and in those cases I combine it with the velocity mutation, which allows you to keep the swiftness boost for most of the level, with boosted crits on every single hit, but it doesn't work on bosses so it's a situationnal build to clear the normal zones.
