No Homunculus Rune

I have a boss cell, but no homunculus rune. Am I misunderstanding something, or is this a bug?

You automatically get it when you beat the game (you can check on the pause screen), press V to use it.

Yes, that is where I have a problem. I have beaten the game, (as far as I'm aware that is the only way to gett the boss cell) but I don't have the rune.

So you beat the hand of the king, killed the king himself, went through the pipes as a green blob and then checked the pause screen to see you didn't have it? (Unlike other runes you don't get a notification for getting this one)

Yes, not only the pause screen, I checked the stats guy, and of course, tried to use it anyway.

I'm mainly concerned if my save file is locked in this, or if it was just a one time bug, or in fact, I did something wrong and it's all fine the next time I beat the game.

EDIT: Now that I think about it, I don't explicitly recall killing the king. I definetly killed the hand. Is it possible that I somehow killed the hand, got the boss cell, and missed the rune before finishing my run?

You need to kill the king in order to get the rune, but you also need to kill the king to restart the run so... IDK
Try finishing the game again and see if the king is there or not.

How you can finish the game on 0BC for the first time without ending aniation?? I don't recall anything like that. It's automatic (animation starts).

you can't escape with success after the battle with Hand of The King if you don't detach your head to finish your run.

I managed to get to the Hand again (sadly I died). The King was on the throne. My only guess is that I managed to kill the Hand, got the cell, and died at the same time? I guess? I can't recall exactly what happened.
