Strange symbol on floor? reacts when i stomp on it but does nothing

So i've found a few symbols on the floor and if i jump and stomp on them they glow red for a few seconds then stop, i've tried spam stomping, hitting it, throwing bombs at it, they just won't open or do anything, anyone know what i'm meant to do?

You can stomp on them to break them, but it works only after you've obtained the Ram Rune. The runes are a metroidvania-ish progression element that open up pathways which you couldn't access before. Once you've obtained a rune, it will stay on the respective save slot forever.
