How do you enable boss cells?

Do you need to beat the game first? I have been stuck on the last boss.

You need to beat the boss first, each time you beat the boss you unlock the Boss Stem Cell, use the cell in the jar room and increase the difficulty then you when you kill the boss on that difficulty you unlock the second cell, each cell unlocks on the next difficulty till you have 4 cells and it unlocks Nightmare difficulty.

So I need to be the last boss and get his cell then? It isn from the other random bosses?

No, only the final boss. Once you defeat him you can activate 1 stem cell difficulty (or stay on 0, you can keep switching difficulties as you please). If you want to go on 2 stem cells difficulty you need to beat final boss on 1 cell and so on - beat him on your higest possible difficulty to unlock next.

Odd, I've killed the Hand of the King three times in a row and he only dropped one Boss Cell.

You need to be playing on the highest diff available for him to drom the next one. The 5th one isn't for killing HotK, but Giant instead.

LOL. Of course. Beating 0BC, activates first stem cell, finish game with it, and 2nd will be dropped. And so on.

Otherwise, there would be just two difficulty levels...
