Quest and Wasteland Exploration
What's the dweller's specialty that important for quest and wasteland exploration so they can survive longer?

Endurance for survival. The higher your endurance is each time you level the higher your health is. Perception gives you a higher chance of finding wasteland quests and during quests perception decreases the movement speed of the cross hairs for scoring a critical hit. Agility increases your rate of weapon fire during quests and luck lets you find more items, more higher quality items, and the ability to make critical hits more often. Additionally the longer you are out exploring the higher the chance of finding rare or legendary items but the longer you explore the more unhappy your dweller becomes. You need to strike a balance.

Endurance for hitpoints and radiation resistance, perception increases your odds of finding a location, and luck increases the number and quality of loot.

Basically, each stat is important. Ideally, you will max out every stat before sending them out. See this page for a description of what the stats do.

Do these things also apply in the shelter too?

Most the facts about stats apply in the vault as well. I mean....you dwellers aren't looking for special places (quests) in the vault or loot so....those things don't matter really. Luck does affect success on rushing productions though.....and supposedly can give a slight boost on caps when you get those collecting production in a room while being close to maxed on that resource. For example, you collect power but, you were already close to max....it maxes out and the remainder converts to extra caps. The extra caps are supposedly increased if the workers have higher Luck values. So, really....there's only 3 'important' stats for production room workers...... Endurance--to increase max HP values, .......the stat related to that production, ......and Luck--for above mentioned reason. However......Agility helps with combat so, doesn't hurt to increase their agilities too......for when the room is invaded by enemy.

Yes. Endurance increases your dweller's health every time they level up. Getting the endurance later doesn't help. Endurance also helps in the Nuka cola bottler.

Strength helps produce power if the dweller is in a power station and helps with attack and defense..
Perception helps produce water.
Agility helps produce food and increases fire rate and chance to escape during raids.
Charisma helps attract civilized people when broadcasting to the waste lands and helps producing children.
Intelligence helps produce stim packs and radaway.
Luck increases the chances of receiving caps when you complete a production or successfully rush and reduces the chances of a rush failure.

Weapons and armor rooms have their production speed increased by placing crafters with the appropriate stat in the weapons/armor room. If you are making an armor that has the highest stat for strength then dwellers with high strength will reduce the time it takes to make that armor, For weapons (and armor with equal stats) you can begin crafting and then click on the crafting room and the info box will display which stat will help speed up production.

If you look at that page, you will see a table with three columns. The first column tells you what the stat does in the vault, so for example, strength benefits power generating rooms. It does this by decreasing the time it takes before the room is ready to be collected. The second column says general gameplay effects. I think they meant when the dwellers are exploring. For example, Luck increases the number of caps you find. The third column tells you what it does on quests. For example, agility increases how fast you fire.
