3 nucacola room (tier 3 with +7end wear) is enough for 200 people.
2+1 (med+science with full crew) enough for self sufficient wault, but if you dont send people to the wasteland. if you send people to the wasteland you need more than 5 medbay. (9*5=45 barely enough to support 2 wasteland wanderer). so you need more empty medbays for storage. same applies to the science center too. so 4+2 total in your work is seems a bit low end.
i suggest you fill all the 2 wide rooms space with med+science.
garden will do no good when you pick 3 nucacola. waste of space so replace with nuclear power. 4 is more than enough for your work woult. but empy power rooms doesnt require anything (not even power to sustain), i allways comfortable with extra power storage.
some says water room is required (to avoid unexplained radiation on dweller). but you will not see any difference. so i may suggest you delete or replace with something else.
radioroom is only required to raise happyness level in your vault. no difference if it levelled or not, and no difference if it used as combined room or not. only differs is total charisma amount in theese rooms. (works with 10s ) no need to use more than 3. but 4 would be ok too.
barbershop, themeshop, training rooms, all do nothing when you done work with them. so no need to use space with them. use as temporary room. and delete afterwards, when you complete work on them.
weapon and outfit shops, required at least 2 of them. when i have steady supply (with 25 wasteland dwellers and 3 overseer quest teams, you can do with less than these amount. but more the merrier) i sometimes worked with 4 or 5 weapon workshop to produce faster. (in time your legendary junk will stockpile. weather you want or not. ie. i completed my leg. weapon and outfits and my junk become 500 pieces for each type. i use all the space with storage rooms)
storage rooms is only dependant how much you are hoarder. i suggest you keep them in a secondary block seperated from main block with empty floor. (let the in vault events like fire radroachs etc starts and ends in that seperate block)

The advantage is that fire, radroach, and molerat incidents won't spread to adjacent rooms. But the disadvantages are that (a) it won't help stopping radscorpion incidents, (b) it takes longer for dwellers to move to an adjacent room in case of an emergency, and (c) you waste space in the vault, risking running out of space late in the game.
This layout helps the most earlier in the game, when you don't yet have radscorpion attacks and you have a lot of empty rooms. When an incident occurs in an empty room, you can usually just ignore it and wait for it to finish. When you have only one dweller in a high-tier room, it is actually better to move your dweller out and wait for the incident to finish.
It may help in end game too if you still have low-level dwellers and rooms that are not fully occupied. Use the zig-zag layout for at least a portion of your vault just for them.
