No. In the fallout universe, people started out in the vaults, they never went to them.

In the sense that this is not real and considered a simulation, yes. It is cannon.

The whole reason and single point of building all those vaults in the first place was to create controlled environments to conduct social experiments. I guess they never actually believed they would really need the darn thing. In truth, only a handful of vaults were truely viable and they were considered control vaults to eventually compare the rest.
