Default keybindings not working

Have reset to default and still not working. The keyboard is working fine in itself, and can move around using aswd as normal, but once in a loco I can't operate throttle, brake systems etc.

I would check your settings in game. I believe there is a function to turn off keyboard controls while in cab, or only within a distance of the control stand. I assume you mean controlling the locomotive using Keyboard while riding in the cab?

(edit: Look for the difficulty settings for your session. When in the main menu, there is a "Sessions" tab. Find your game save and click on the gear next to the difficulty setting.
