The rotating glyph puzzles have come in 2 varieties for as far as I've come.
1) Just match all glyphs from top to bottom with whatever is displayed nearby.
2) They are some sort of Sudoku puzzle, just make sure that there are no duplicate glyphs on any row or collumn. Start out on the row or column with the most stationary glyphs and figure out which one(s) is/are missing and fill in that blank. Work your way from there.
As far as that gate in the floating room, I haven't figured out to enter it but you can access the room behind the blocked door with the spikes in the middle. Right after your first gylph puzzle in the floating room, you can grapple down just next to the door with the note attached. Once you're down, you're in the room that's behind that blocked door.
Which only leaves the gate with the treasure chest.
Will update if I find a way.
You'll get there a little later when you get to a library of some sorts. It's a "secret" entrance from there.
You can grapply up on both sides, but on the side where the Goblin? is standing on the ceiling, is the doorway to the rest of the Dungeon
To get there, you need to push some of those big blocks out of the way.
If you've made a mess of that puzzle, reset it by whistling the last learned tune on the glowing glyph on the ground.
