Can I cast spells with 2 hander?

There is no limitation for magic use. You can wear\use any armor\shield\weapons. Some equipment can decreas your "action speed", but you cant "fail the spell" (you can only miss - if enemy has a high defense or resistance).

Well, technically, if your action speed takes too long, you are at a higher risk of being interrupted before getting the spell off. I cant remember if this consumes the spell slot or not.

Yeah, interrupts will consume a spell use, the Rapid casting & Combat focus feats are a godsend to prevent this.
Not 100% sure if your initiative affects spell casting, as it's technically a separate action to starting another turn(TB)/action(RTwP), though i think dex can speed it up...PoE stats are weird like that.
