As for cpu vs cpu, the only way is to make a 3-4 player match and you intentionally lose just to watch them.
Keep in mind that the cpu difficulty is mostly based on character than the handicap difficulty meter. So if you were expecting to see the best, you'd typically pick the ones on the bottom of the list over the ones on the top (that's not a hardset rule but merely observation).
Handicap difficulty I don't think affects AI's behavior, just gives them a handicap that would inconvenience (or benefit) a human player
I remember that, in the DS Puyo Puyo games, if you held SELECT while choosing match difficulty, the AI would take control of the field you'd be playing in.
I'm unsure if it's possible on this game given you can't choose difficulty.

How are we supposed to know which character has the best AI?
