No you shouldn't—for a first playthrough anyhow—the director's commentary shuts off the regular voice overs so you won't be able to grasp the story as much.

Agreed. Play it all the way first without the Director's commentary, then play it after with the commentary. The commentary was very interesting. I hadn't noticed the shape of the mountain, so that was an interesting piece of information.

There are a few things that the directors commentary showed me that I didn't know. It was very rewarding, but would have ruined the game the first play through

I just played it for the very first time and had Director's commentary switched on. Couldn't make sense of it. Now I know why. No voice overs.
