If you're using a wrapper like SCP, you have to register two profiles, or both will read from the same input. If you're using legit xbox 360 or xbone ones (or their bootleg equivalent), it should just be plug and play.
It seems pretty stupid to have to remap the Dualshock 4 or any other controller when it already works with Big Picture mode. Is there a guide on how this is supposed to work?
What you should see, at least in 99% of all other fighting games, is a screen that has 1P, 2P, 3P, 4P...so on at the bar at the top, and controllers underneath. Players will then grab a controller, move it to the P slot they want, and there you go.
If it's not setup like that, and I have no way to test out just how Xrd/Rev works locally right now, then it should just be a simple matter of 'device #1 is P1, device #2 is P2.' Like I said, if you're using a wrapper, or other unathorized program to get sorta-semi supported controllers and plastic devices to work, you're gonna have to setup the profiles. Play without steam big picture, for one. It may restrict you to single device and P2 will have to use the keyboard.
If you're using actual steam controllers...then I can't help you, man. Sorry. Anything from wineglass styled american arcade boards all the way up to PS3 bootlegs I can do, but I ain't got a clue about the steam controller or even its software.

You should see the above screen with a pile of 'controllers,' each representing a recognized device, whether it be a steering wheel, a joystick, legit controllers, third party solutions, or whatever. If the game detects them, they'll all be there.
Move the one you want to the player slot you want, then play. Multiple SCP PS3 controllers don't work for me, but one hori fighting commander set to act as an xbox 360 controller, and a keyboard fooled into acting as a PS3 controller running through SCP works fine, as you can see.
If you can't get it work, make sure you actually have the proper drivers installed.
Far from it. Microsoft has a stranglehold on the platform, and only devices running under their protocols are natively supported. If you want a PS3 controller on windows, you have to use a wrapper, which is a program that tricks windows into thinking it's an xbox controller. Dedicated fighting game devices feature hardware switches, so you can flip between windows, xbox, and playstation with a button press.
Makes it easier for pick up and play, as well as tournaments.
For anything PS3, I recommend SCP. It's a non-invasive, no ads, no nagware, no-nonsense power solution, but installing it initially can be tricky. Development ended years ago, but since the PS3 stopped being relevant even longer ago, it doesn't matter. Handles up to 4 controllers, and even does bluetooth if you absolutely want to bother with that.
