Assist Mode can be used or it's like hack?

Assist mode is an actual option in the game, not a hack, doesn't block achievements or anything. You just get a mark of shame on your save file that indicates you turned on Assist Mode. It'll always be there if you turned it on ever. Unless you delete the save.

Assist mode is specifically designed for those who otherwise would be unable to complete the game, physically or mentally.
Some may abuse the system, but legit... why should you care?
It has zero impact on anyone else's experience except their own. It's an entirely only single-player game after all.

assist mode is not a hack or anything of the sort--it's just a handy little tool to help people who can't play the game otherwise (due to the game's difficulty, physical restrictions, or just not feeling like doing all this crazy crap).
you are free to use or not use assist mode as you wish, and if you activate it, you'll get a cute little slip on your save file that disappears if you deactivate assist mode.
nothing is disabled if you activate assist mode (i got a whole bunch of strawberries and the 1UP achievement with assist mode on bc i'm lazy), so you can totally just turn on assist mode and get a golden if you feel so inclined.

Does this means i can disable Assist mode at any time after enabling it? Or it will stay enabled forever on that save? And if i can disable, will the save be "tarnished" because of that(i mean, having a sign i enabled it at all)

Assist mode can disabled whenever, and whenever you have it turned 'on' it'll showcase the assist mode bird logo stamp on the save-file, but it goes away again after disabling assist mode.
