So as for your example, the rogue's "buff party" slot bonus requires a buff spell to be placed in that slot. If you do so, then whenever you trigger that spell, the entire party will receive the benefit.
If however, you choose to put a fireball in there (which is a projectile) you can still use the spell regularly, however you won't get anything from the slot bonus.
It's a minor way of min/max'ing while playing around the RNG in this game. Don't feel too bad about not getting all the slot bonuses, but you could try to play around them in the early game, till you find the specific spells you need later on.
If not, it's always best to slot some spell in there rather than leave them blank.
