Edit: it isnt missable anymore, they moved that to a cave below the thalmor embassy, so with that there are no missable ones.

I've completed this quest once. Not again, no.
You can miss one, the one in the Dark Brotherhood is unattainable once the Falkreath Sanctuary becomes inaccessible. That's the only one that is missable.
Looking at the wiki, that isn't true. Entering the Sanctuary after it gets destroyed can cause the game to freeze, but the official patch is supposed to fix that. There is no other indication that that particular stone is missible. There was only one missable stone (the one at the Embassy), and that got moved so its no longer missable.
Riften is of course under the thieves guild so has no special mission.
And of course you need to have done fifty/sixty missions (not counting special missions) to unlock the leadership of the thieves guild.... which is just another set of armour with better bonuses.
You also get Amulet of Articulation when you become guildmaster.
You don't need to do that many, doing all 4 special jobs is all you need to do. If you've done that you can become Guild Master at the conclusion on the questline. You could become GM in a little as 24 jobs. You get extra stuff added to the Guild Masters little area for doing more missions, but it's not required.
