Isran won't talk to me
Not gonna state the reason cause spoilers...do I have to revert back for him to speak to me?

Isran will not work with Vampires, you can still be a werewolf if you wish he doesn't seem to mind that. And you can become a Vampire Lord later on after Dawnguard is done through Serana.

He does not talk with you anymore or just don't talk? I am curious at which point of the Dawnguard questline you are, the solution may be dependent of it.

Well let's just say I defeated some guy who's part of a race thought long gone and the remaining survivors are horribly mutated. That's how far I've made it into the Dawnguard questline and I want to get help from Isran for another battle.

When you say revert back, do you mean revert back from being a vampire, or revert back to a previous save? If you advance the quest too far as a vampire, it may be too late, so yes you would need to revert back to a previous save. If Serana still has the Scroll, then it may still be possible. (You can cure yourself in Morthal.) I wanted to test some things as a vampire (in Legendary Edition) and got thoroughly horked by being a vampire, because the vampire exploits are "fixed" in my version. Absolutely no value there. Reset from previous save.

While you may be able to get the exploits in SSE, if you want to do all of the elder scroll stuff with Serana, you may have to wait until after that is completed before you make your "decision."

I got the quest to cure myself as a vampire shortly after coming back to Fort Dawnguard after I became a vampire to do a previous Dawnguard quest. Well what do you think I'm referring to lol? I guess my only choice is to do that quest in Morthal then.
