And SKSE is not baked into the Skyrim SE 1.6.353 Anniversary DLC you'd have to download it here SKSE.silverlock.org
and would need this version:
Current Aniversary Edition build 2.1.5 (game version 1.6.353)
Are you using a mod manager and if so which one?
I went to the page you gave before and downloaded the SKSE.
When I open the MODS page while in the game, I did a search for SKYUI and nothing.
What am I missing here...

personally I wouldn't (and don't) bother with the in-game mods section period

But are you talking about Vortex or NMM?
I was using NMM but its all configed for Vanilla.
So, I installed Vortex and then the SkyUI and it worked.
HOWEVER, now all the mods I activate within the game itself by clicking on Mods, those all get dissabled! So now, It appears I can only have one or the other.
Active and working SkyUI or all the other mods actiavted in game.
I'm starting the game with the latest SKSE as posted above.
Mods obtained from the mods menu can be imported into Vortex though.
