When should i start playing them?

You can play it on beginning the Game. You should play the DLC's before you start the Main Story. All Enemies in the game scales on your Char Level, so if you start with Level 1, all Bosses etc. are Level 1(DLC Bosses too)

But you become in the DLC's 2 new Partners and 2 new Classes.

The new Partners and Classes can you use in the Main Story too. But you must play first the DLC's before you get this.

So if you first play the Main Story, then you cant doing anything with this partners. Because you havent quests
