My advice? Go onto the NG+ that just unlocked for you, Way of the Strong. Do the side-mission in the first area where you go around with Tachibana-waifu looking for treasure. Let her do the heavy lifting - she'll kill most things handily, and if you're in there gathering attention and blocking/dodging while she makes the kills, you'll make a heck of a lot more Amrita than grinding Queen's Eye over and over again (that's a terrible level for grinding in any case; Marobashi or Hyakki Oyogi are both easier and yield far greater rewards).
Once you've hit about 150 or so, or whenever you feel comfortable, jump into the first DLC. I will warn you, that first mission is a pretty big spike in difficulty, and is quite the skill gate to get you used to the rest of the game.
I'll also say that you've just scratched the surface of what Nioh has to offer. To my mind, there have always been three games in Nioh; the first playthrough, which you've just finished the base game portion of, and then the NG+ cycles up to Way of the Nioh (NG++++), and then finally the Abyss (which is a hell of a lot more fun than it sounds at first, especially if you're able to do it with a good coop partner).
Nioh is a deep game with a lot to go through. If you're willing, it will be a rewarding experience for a good long time to come.
If you're unwilling to put in the time, however, you might be better served taking on Nioh 2. Who knows, the DLC for that game might be coming out about the time you're finishing your base playthrough there, and you'll be looking down this exact same decision again ^_^
Yeah, most of the side missions in the 6th region are dual bosses. This is not always the case - remember that at release, this was the end game for Nioh, and thus the big challenges were put here.
Heck, a few of these like Return of the Gourd are significantly harder than a good number of the DLC missions. Don't feel the need to complete everything here before you go forward; as long as you have Marobashi unlocked, you are safe to happily proceed.
