The other hard boss fight for me was the Ogress. It took me a while to figure out that you can hug her real tight and not be hit by her swiping nails. I even went to the effort and exchanged my odachi for a newly forged katana for lady Ieyasu.
She immediately became manageable after that, but she felt like a real show-stopper those first three or four tries.
Hardest main game boss: Tachibana Muneshige
Hardest DLC boss: Sanada Yukimura

+1. OP ought to have asked which are the easiest boss(es) b/c they are only handful
I'm an average player, but i think that changing the angle and the translation of the polygons during an attack (say, at 25/30% of the animation) is a cheap way to create difficulty.
Kind of stupid that yokai bosses can't 1-shot me but a human tap me with a broom and I die instantly. :/
The worst combo I could think would be Umi-Bozu and Giant Frog. One highly mobile with a nasty long game and one almost immobile with titanic AoE and nasty OHKO.
I just met Tachibana and... It was not very easy, but few tries and I managed to kill him without summoning anybody! :)
Just use pillars (to hide) to drain his stamina/ki ;) And change stance to high from middle (I used Dragon Sword).
Yokai Bosses vere harder for me.

In Dark Souls 3 one Boss (some other too but was not so easy) was in room with pillars too and it was one of Bosses I killed first try! ;)
