You don't need to use your both hands though on standard keyboard as both of the ctrl-buttons work, so you can play it only using the arrow keys and the right ctrl-button. I don't know how it works with XBOX-controller as I own a PS3 myself, but in the options it states that a controller could be used.
For your question about OSK, there were no settings in the game that would allow it to be in windowed mode, and I don't know of any modifications that would allow it to be so. So using the OSK seems not to be an option for this game.

I found out that you actually CAN play this game in windowed mode, it just does not appear in the settings. Used Google and found out how to get it into windowed mode - and you actually can use the OSK for playing this game! Just do the following;
"Q: I want to change the resolution or run in windowed mode
A: Limbo is carefully designed for fullscreen 720p so this is the recommended setting. However as some people really like to tweak these things we have added an option in the settings.txt to change backbuffer size and a setting for running in windowed mode. To run in windowed mode change “windowedmode = false” to “windowedmode = true”
And there you have it! Just do that after you've installed the game, and you can play it in windowed mode.
