Whats up with the eye poke scene?
Why was it necessary for the story? Isaac just needs to destroy the marker, right?

It's just part of the story, so try not to make too much sense of plot.

The scene represents Isaac's release with his inner peace that Nicole is real when she haunts him and that they are whole now.

I wander why his girlfriend's illusion can kill Isaac himself? She has already dead but tried to kill Isaac twice :|

in one scene where she is trying to kill you with a syringe needle when you shake it off, you realize how Isaac is holding the needle himself, so its not like she is real, more like she is trying to take control of his mind and make him hurt himself.

thats why in the end of you fail he kills himself with the harpoon gun

I always assumed the eye poke machine was a necessary step in destroying the marker. Gotta follow the steps. Like perhaps Isaac needed to use it again to find out what he needed to do inside his head? But yeah who knows lol

The eye poke machine unlocks the deepest part of the brain where all the marker information lies which is why you need it. Basically, that's what shows you how to destroy the marker

I got a pretty good answer for you to answer all of these dead space questions, becuse i did my research:

1. Nicole trying to kill you: This may come to a shocker to you but... thats not Nicole. The Nicole you see and hear throughtout the story is the markers way of trying to control Issac. The Nicole is a part of the marker that was trying to lure Issac (and Strauss) close enough to consume or "make us whole." The marker needed the makers to do so, because if you follow the story these markers are a sort of "beacon" or tower to send a signal for a giant alien moon monster to come and consume the planet or whatever the current marker is located on! Does that help about the whole "Nicole" situation?

2. The eye poke scene: Ahh yes... that scene. One of the most strange and confusing parts of the dead space 2 story, allow me to explain. That machine was used for the patients like Issac and Strauus to find out how to create and mass produce these markers. The machine helps unlock and show the part of the brain where the infromation is stored. Once the needle goes in whoever is operating it outside of the machine has access to the patients memories. With the memories and infromation (marker codes and data) they can use this to create markers. Side effect of unlocking the stored info on the patient may cause hysteria or visions of dead family/loved ones.

Some more lore: The markers are like i said just the thing that start all of this. The moons created the origional marker on the planet in the first dead space. But since the planet froze up it stopped the infection to possibly spread. But in come the curious humans, drill a hole in the planet and find the giant monster that was with the marker. But as seen in Dead Space 2 humans found a way to recreate this by finding the "Ishimura" with the marker on it. They did some tests and found out it puts images in peoples head and makes them hear/see things (just like how Issac was going insane throughout the story).
