Do shrines reset?

Or are they single use only? I'm a new player and have come across these Shrines (like of Experience and of Mastery) but I'm reluctant to activate them if I can only use them once - I'd rather wait until I really need the buff, if the shrines are single-use only

I can't remember if they do at the next level, but I know its one time for the first level.

AFAIK they don't ever reset within a run... ...but when you reenter the map, sometimes new ones pop up in previously unused shrine spawn spots...

Each individual shrine is one time use only. When you use them you cannot use them again in that game session. Multiple shrines of the same type spawn in the game. Using one shrine of mastery, for example, does not prevent you from using another shrine of mastery elsewhere. You just won't be able to active the original shrine again.

They get reset/randomized along with chests and monsters when you start a new game session (exit to main menu, start new game with that character or start new game from desktop)

FYI, the buff from the shrines is very short term. You might as well just use the shrine when you see them.
