There's an achievement for completing normal with only one mastery. After normal difficulty it should just be a self-imposed challenge. Completing normal with one mastery isn't very hard at all, you can play whatever mastery you want.
General drawbacks of a single mastery build are worse stats (due to missing 32-40 mastery points spent in your second mastery), fewer build options and diversity, you'll have more skill points than stuff to spend it on long term, etc.) Every mastery should be viable for single-mastery legendary clears I think through I've only done completed a full run with Warfare single mastery. Masteries that are strong by themselves make good options (Warfare, Hunting, Dream, etc.)
Usually I play Conqueror. The only thing I miss from Warfare is Onslaught.
I tried playing with Warfare only. I feel like I miss adrenaline+rally all the time plus all the shield active and passive skills. Very hard for me to complete with pure warfare dual wield character. A lot of running away, potions, charging with War Wind again and so on. I cannot just smash into enemies and simply brutally slaughter them in a few hits/shield hits.
I actually feel no handicap with playing defense alone. I make better use of the limited skills I have instead of trying to use a piano build.

Though I do usually try to keep my pets healed, and of course trying to avoid getting hit myself.
