Are you still unable to speak in Discord when the game is running?
If so, please give the following steps a try;
1. Ensure Discord has Mic permissions;
Open your Windows Settings > Go to Privacy > Microphone
Go to the section of Allow access to the microphone on this device, click Change and turn the option on.
Also, make sure the option of Allow apps to access your microphone is enabled and Discord is ticked.
2. Disable the Discord Overlay
Discord > Settings > Game Overlay > Untick 'Enable in-game Overlay'
3. Ensure Discord is running as an Administrator
