Is there a second time I will be riding the beast in the game? I am 9 kills away from the achievement, and I do not want to have to start a new game just for 9 kills...

You have to replay, there is no second time. Also you should save before trying to get the achievement again just in case.

does it have to be in one straight run? I would have had it i think if i wasnt trying to understand that it could lock on at the beginning with the charged attack

Yes, you have to get the achievement in one run.

I can confirm that you DO NOT have to get this in one go. You can simply just reload the quick save in the middle of the fight, right before the small green bats spawn. I got the achievement on my first run today by reloading a couple times.
