Should I read the comics before playing this game?
The game looks fun, but if it's necessary for me to know about the universe before hand I probably won't pick it up.
Does this game serve as a good entry point for those unfamilliar with the comics?

It is not necessary to read the comics to enjoy the game’s story or to become familiar with the characters! People who did read the comics will have the advantage of knowing the characters and their background, and how they came to be here. But we made an effort to introduce the characters to players, and provide interactions throughout the game that give you a better sense of who they are.

I had no problem enjoing the story, after playing, infact I will probally buy the comics now.

You can get reprints in TPB format or scans through sites like Comixology.

I played without reading the comics. Then I read the comics afterward... and, nothing changed really. All I learned is that Garrison has a personal reason not to use his sealed-away sword, and Gully's past with her gloves. (They were her dad's.)

The comic kind of finished on a cliffhanger so you won't be missing anything. If anything, you will feel really weird after reading the comics because you'd think that the game continues after the comics... It doesn't.
