How I unlocked every 20 characters

1. completed tutorial 2 times (1st time for character, and 2nd for achievement)
2. first completed story mode selecting left character every time when it asks before battle
3. completed arcade mode on normal with every character which is needed to unlock all achievements for robo, family, hall of fame, girls power. during this also made all other achievements like for big blocks popping, except achievements for online mode play and 5-chain pop
after that i only had Thompson locked.
4. played survival mode for unlocking Thompson (atleast 10 opponents needed to defeat if i heard correct - i got 12, so Thompson unlocked)

i got him during my several arcade completions which i mentioned, so dont know what is his unlock condition, but probably nothing special.

Johnny Turbo → Clear the first tutorialPlay
Blackjack → ArcadePlay
Thompson → Survival
Isaac → Clear the story (zombie route)
Jim Akji Volos Elise → Defeat in story mode (possible in the first week)
Hunter → Clear the story with quotes (including tags)
Kawase ... where?
