no, if you miss something you will get another chance later.

a few achievements are mutually exclusive and thus will require a 2nd save file to get.

you can only get the community center or joja achievement on a single save so if you get one you need a different save to get the other

Yeah, there one achievement that you will have to play the game a second time for, because you have to make a choice that will make the other one inaccessible.

This may be somehow avoided, if you collect enough money, go to sleep to save the game, make a save file backup, purchase all Joja upgrades, unlock achievement then restore save file...
