For co-op, all players must be online to play together. But you can play even if one of your players isn't there - their character will just join in next time (so in a multiplayer game you might go to bed in Spring, and then next time you play the game might be up to Winter already!).
Only the host needs to be online for the game to run, and if the host plays the multiplayer game by themselves (with no other players logged into their characters) it works just like single player.
The biggest difference between single player and multiplayer is that in single player time pauses when you talk to people or are in a cutscene - in multiplayer time still passes. So there are times when you might end a cutscene and immediately pass out because it's after 2am.
