I know that you can find dust sprite in caves. Can I find it in Mr.Qi's cave(I don't know how to call it in English.)?

I don't think I've seen any in the hard mode of the mines, no, but I've skipped a lot of levels (mainly those with vomit ghosts) with staircases.

The cave changed by doing 'Danger in The Deep' quest from Mr. Qi is generally called "dangerous" levels. Some may say "Shrine of Challenge" cave or Mr. Qi's cave.

From Special Orders quest from villagers, you may receive Clint's quest to slay one type of monsters from either Bat/Dust Spirit/Skeleton/Grub.

I had the same quest, and if I do recall it correctly, Dust Spirits in "dangerous" levels is equivalent to mushroom monster (it appears no official separate name is given, it is still called 'Dust Spirit') which spawns in floor levels between 40-69.
