Also, you need to first have a basic coop - you then get the option in the build buildings menu to upgrade the basic coop to a big coop then to a deluxe coop. Same rules apply for barns.
Make sure you have enough money and materials for each building/upgrade. Materials need to be in your backpack, not a chest on your farm.
As for animals, no your coop/barn does not need to be full. It can be upgraded without having any animals in them, which is good because this makes it easier really.
I had a basic coop which I then upgraded to a big coop. I also have a barn, but I haven't upgraded it at all.
I always carry around wood and stone, but even if I don't have enough, I should still be able to see the deluxe coop listed in the shop.
* EDITED REPLY: If you have no scheduled house upgrades (as detailed above), enough funds & materials and you are still unable to access the buildings menu at Robin's shop to find the building upgrade you want (in this case the deluxe coop), are you trying to do this on a festival/event day? (that sometimes prevents shops from trading as usual - but you may still see Robin in her shop or Marnie at her farm).
I would add again to check your inventory just to be sure - the game can be very fussy as to the exact type of wood, stone etc. needed for each building/upgrade.
If none of this works, you may need to either wait 3 in - game days (go to sleep, not pass out) to see if it sorts itself out or start a new save file.
Are you using mods or playing the vanilla? If it's the Vanilla I'd say try asking Concerned Ape on Twitter or the community wiki. If you are using mods - take a look at your SMAPI logs - anything in red is a problem which needs fixing. The log should detail the error and which mod is causing it, from there you can contact the mod's devs on Nexus with a copy of the SMAPI log for them to look at/fix. You can always use the SDV mod community boards to post a querry including the log and hopefully the friendly and helpful modders on there can help you fix the issue.
