Not particularly.
Like all fish, as long as you're fishing in the right season/location/time/weather, it can bite. Then it's just a matter of catching it.

If you miss any of them you can't catch a puffer fish. Be aware that puffer fish has the shortest appearing duration in a single day(4 hours), you may want to use the time spot glitch while fishing.
+2 or +3 Fishing food and a Cork Bobber might help.
Ah, yes. If you don't have that one, then it's not an option.
+3 Fishing adds a whole lot of bar length all on its own, though. This is why everyone thinks Fishing is hard (bar's tiny at low levels) but also why high-level fishing is boring (bar's huge).
Correct, though that will not count as you having "caught" it.
(Though yes, obviously that doesn't add it to your fishing collection).
