When you FINISH the regular run, and want to start NG+ it will ask you to choose a save slot that it will base its "stuff" from. That's why if you don't have a finished game (you must finish completely first) you can't really use this feature, and why if you use a craptastic run for it, you'll get only what few things you found on that run in addition to your play through.
You can do NG+ as many times as you want, I haven't *tried* using an ng+ slot but I don't think you can build on it? (someone probably has tried, and would know, please correct me if I'm wrong on that.) But I'd say make sure that you have a full run saved - even if you have to MANUALLY copy the file and reinsert it later, over another save file.
I have to say that NG+ using Corvo with some Emily powers (namely Domino and Far Reach even if it duplicates your teleport with corvo's blink - the Pull power is essential), is a TON of fun. I just outright prefer Corvo, so having access to a couple of the newer powers along with the older more familiar ones, was excellent.
