You have to aim the head.
Head shot = instant sleep

Yes there is a blueprint but not sure where is tho, might be randomised like bonecharms

you'll find the combat sleep dart formula in the 3rd mission, near the entrance to the asylum.
here a vid with the exact location:

The thing is, I somehow managed to miss it on my current PT (and I explore a lot, probably even more than during the first run). I have a strange feeling that some of the blueprints that I'd found on my first run are not where they used to be and\or different.

E.g. a nook with the howlers behind the station (before riding to Jindosh) - I remember one in a dresser, right behind the lady. I remember it being there because it took me some time first time, but this time the dresser was empty (a rune, aoins and darts - but no blueprints).

Can anyone comment on that? Are they randomized? Could it be a glitch?

Um. In DH 1 that also meant there was a chance to kill a target if you shot them in the head with a sleep dart.

There is one, yes. It's known as 'Combat Sleep Dart' because that's presumably when you use it.
Personally I don't like it; the delayed version lets you start controlling other nearby enemies while it takes effect, or escape for cover before it does so.
With instant, everybody notices your first target go down right away, and you lack the time to react effectively.

Ok, this time I got it from Hypatia on the ship, before she left. As I've mentioned - I had missed the upgrade during the Addermire level. It's like the game gives player the second chances with the missed (crucial) blueprints.

yes, if you miss a blueprint, you can get it in the next mission.
