how do I equip new runes ones all the slots are full?

I don't think that that is possible - unless you can add another slot at one of those shrines that adds slots. But that, too, is limited.

Well I mean you can remove the runes from the slot I just don't know how to drag and drop new ones into the slots

I don't think that you can. Once the runes are put in there, they'll stay there.

That doesn't make sense why would you be able to remove runes then? and theres 3 runes per element what would be the point.

Ah ... I see. I'm sorry, I misunderstood what you asked about. You meant spells in your quickbar - not talismans in the slots of your items.
You have to open your spellbook - either by pressing the hotkey or by clicking on your spellbook in your inventory - right next to your character on the left.
Then you have to select the desired school and spell and simply drag it over into your quickbar - just like you do when you equip weapons.
