Players who are more familiar with platformers most likely can finish the game faster. The Definite Edition added a few hours worth of optional content and also quick travel which generally speaking makes the DE shorter because backtracking is faster.
And of course you can complete it one run... maybe you mean in one sitting? I mean, people speedrun this game and there's an achievement for finishing under 3 hours, so yes it would be possible... not on your first run though, you gotta know what you're doing first.
If you mean "how long will it take for first playthrough in average", I would say 10h. Personally I used 20h.

for a later playthrough : the usual player should take a bit less than 3 hours. My record was sub 2 hours when doing One Life Mode
