Transmute is very useful for saving useful items of various kinds... unless you're trying to do the achievement where you don't return to town ever, of course. That would be the exception, otherwise points aren't wasted there in my opinion.
Mana regen is a very wise move, and health regen can be good when you don't have what you need for the mana regen (since it saves mana you might have used to heal).
You might just want to put all points you can into beefing up your character's defences (your magic offence is probably the next best thing to put points in every now and then if you're feeling okay otherwise). If there are hearts on the way to something else you'd like to have, you can get those. Also, don't ignore crafting and potions-- they're a huge help to survivability since you can craft potions anywhere and they can make materials you'd never use for your equipment suddenly become very useful. That being said, although I am a glass cannon fan in other games, I haven't tried it for this one since it's so risky once you get past the early stages.
The character i'm playing is mainly magic based. I have a striker, about 200 armor, and 200 magic now. Sitting on nearly 700hp, those undead vikings deal the most damage at about 60hp per hit to me, but i just slay em with 2 quick light magic attacks, for 14mana, kill for 13, so 1 mana per kill, and i usually dont get hit.
This build im using is working for me, i can heal 200hp with just 20 mana and i have 460mp.
I usually sell most materials, if their not mp regen, mp restore or hp restore, i sell them, i use mp regen to forge and mp/hp to alchemy.
Im just now branching down the alchemy/enchant/item chance tree now.
I also got telekenisis so i can steal chests if i dont want to fight an army.
(And I'll probably also go for a glass cannon build myself soon, once I can get back around to this game!)
I started an alt, on the labyrinth mode, got stuck at floor 11, due to the insane difficulty ramp, this character went with the combat tree, was faster to level, but relied on potions and outclassing defense to negate all damage.
So i'm going to create 1 more character, highest starting attack damage, go full combat, get the skills for heal then full explorations tree.
Ideally always start at floor zero to get as many gem upgrades as possible.
I think the endgame & NG+ requires a decent amount of gem acquisition. Relying on atleast 100-150 attack, 100-150 defense, 1000+ hp from gems alone for the endgame content and preparedness for NG+, without farming those gems, THEN the game feels unfair.
