Of course you may be right, but your explanation could be used to explain a lot of things.
Doesn't is specify that "she died recently"?

- Her hair similar as the wife we find in the lab in the Painful
- She saves Buddy twice
Though there is something I dont understand. At the very end she still fought Buddy. Is the Yado's trumpet control THAT specific ? Why would he do that to her anyways since he wants her to help him rule... I dont get it.
I do believe sweetheart is female, but that doesn't necessarily mean it's her mother. I think it was Yado ordering her to save Buddy moreso than her just saving Buddy on her own.
And Yado doesn't want Buddy to help him rule, indeed he was expecting her to either die or turn into a joy abomination at the end of it all. He just wanted her to help clear the world of regular, non-joy mutated humans. That way he could just control his abominations in peace (we do know the majority of the leaders we kill were killing off the mutants).
You have a point, though all of the other mutants have "normal" names, like Harry, Louis, etc... It would be very strange to have THAT mutant called sweetheart just because its female. Now that I think of it, Yado mentionned a daughter called Nancy, even though people seem to believe Nancy is Buddy... To be sure about the identity of Sweetheart we would have to be sure of Buddy's identity IMO. Because I wouldnt see why he would call someone Sweetheart if its not a close person
I do agree your points make it ambiguous though.
Your theory works as well, he calls them children. thought since he doesnt look very sentimental about his wife and daughter, but still calling them children doesnt stop him from piling them up and sitting on top, so ... not sure about this
