In general, Pro Motion is for creating/drawing pixel art and animations by hand drawing stuff.
Spriter lets you use images you already acquired or drew in different programs to create animations.
They both could be used with RPG Maker, especially Spriter with the RPG Heroes art pack.
Both software you can use though if you want to export as normal bitmap images to use as sprites in your game (this is a standard workflow) but the benefits of having native support for Spine files inside GameMaker Studio 2 is great!
