As for Bloody Palace, it unlocks after you've beaten the game in Son of Sparda difficulty mode with any character.
As for "Super whoever" you have to beat Must Die mode with each respective character. Trish/Lady don't get "Super" unlocks.
The difficulty modes, in order of difficulty are: Human > Devil Hunter > Son of Sparda > (Dante) Must Die > Legendary Dark Knight.
Then you have Heaven & Hell, where everybody dies in one hit including you, and Hell & Hell, which is basically Son of Sparda mode but you die in one hit. In both of these modes, you are given 3 free Gold Orbs so really you can die up to 3 times before needing to use a Continue.
Bloody Palace unlocks after beating Devil Hunter (normal) with any character.
Super Nero and Super Dante are unlocked by beating the Dante Must Die difficulty in Nero/Dante mode, while Super Vergil is unlocked by beating the same difficulty in Vergil mode.
EX colors are unlocked by clearing the minigame during the credits. In the Nero/Dante mode, this means protecting Kyrie from enemies until the timer reaches 0, not letting her get hit once. In the Vergil and Lady/Trish modes, you don't have to protect anyone, and instead just have to kill enough demons during the credits. If successful, the EX colors are unlocked for whatever characters you're playing
The difficulties go Human (easy) > Devil Hunter (normal) > Son of Sparda (hard) > Dante Must Die (hardest).
As you climb up the difficulty ladder, enemies start having more HP, dealing more damage to you, and even using new attacks they haven't used before. In addition, on Dante Must Die mode only, enemies have the ability to activate their own Devil Trigger if you take too long killing them.
In addition, you have three "gimmick modes" with special rules.
Legendary Dark Knight is an extra gimmick mode that is set to the Son of Sparda difficulty but has ridiculous, immense amounts of enemies spawn every encounter, usually filling up almost the entire screen.
Heaven & Hell is another extra gimmick mode where everyone dies in one hit, while in Hell & Hell mode, only the player character dies in one hit, while enemies are set to Son of Sparda difficulty.
All of these modes can be unlocked without buying any extra dlc. As you clear modes, more unlock.
As for the rest of the dlc you can buy, red orbs and proud souls are just in-game currency used to buy items and skills respectively, you gain enough during normal play so there's never any real need to grind or buy these dlcs to get more. Blue orbs increase your maximum HP, but your HP bar can be maxed out without ever buying them. To do this, you need to buy every blue orb in the in-game item shop, find all the pieces in the missions, and clear all 12 secret missions hidden in the world.
From what I hear (haven't played the original DMC4), Special Edition gives players more proud souls than the original, so buying proud souls is more unnecessary in this version.
(That's specially good if you're starting and want some quick proud souls)
