It improves your throughput to have the lumber already cut for the builders.
Go ahead and try playing without a lumber shack or miner and see how things go. It won't be pretty.

U will notice builder cut tree slower then lumber jack
Fletchers/Tumblers (and all other refining/manufacturing jobs) actually can't harvest on their own, but that can grab stray resources on the ground. So if you don't have the harvesters a large majority of your refining/manufacturing operations will mostly grind to a halt. :)
The only workers who can harvest are builders, harvesters and labor golems. But the builders will only harvest when the village is completely out of the resource they need for a project.
Adding on to what others said above, harvesters also do their work slightly faster, and get additional speed boosts once you get a tool smith cranking out axes and pickaxes. :)
But at corruption containment phase. It best builder harvest themselve as it far from base. Do it depends.
