First, make sure you have the skill Charge Ki equipped, and if you're using a gamepad, it should be Right trigger + whatever button you assigned it. On KB&M, I have no idea...

In this game, by default, you gain KI by attacking in melee. To charge up you must buy the skill "Charge Ki" from the Skill Shop in the Industrial section of Toki Toki City. Then you press the button combo you assigned it to and bam, charging!
