DST has its own seafaring content that to my knowledge has never been added to the original. It has more stuff to craft, more biomes, more bosses, more QoL and I believe better UI too unless the improved crafting menu has been ported over into the original.
Both games have a story of sorts, and DST is more advanced lore-wise. But this should have next to no impact on your general experience unless we're talking "end" bosses or how a few things look or how they are being commented on by the various characters upon examination. The story/lore generally in both games is more of a faint background noise and not very relevant for the gameplay at hand.
DS has this "adventure" mode which as far as I played it tells the original story by making you change worlds every now and and which has some precrafted interlude levels of sorts, apparently. But I don't think that's much to write home about, maybe you can check out that stuff easily on YouTube without having yourself to go through all the survival and open world stuff surrounding it. I switched worlds only once in adventure mode and went back to playing DST again, but maybe that's just me being impatient and spoiled by DST.
Some biome setpieces and items never made it into DST and are only exclusively found in DS. But that too is a rather small thing compared to the rest I mentioned. The reasons for their absence in DST is mostly griefing-related.
Some characters should still work a bit differently in DS too. The whole "insight" system (skill trees for characters) and reskinning characters, items and structures only exist in DST. So if you love to customize stuff then you totally should pick DST.
If you choose to play DST solo then maybe you want to get a mod that adjusts the health of bosses accordingly. They are more beefier in DST and also work slightly differently to their DS counterparts - or at least those that exist in DS. Because again, DST has more bosses.
Oh, and DST has endless mode. In DS and in the survival mode of DST, as soon as you die the world will reset. If you want to play solo without having to start over when screwing up then DST has that endless mode for you. But mods for DS exist that let you continue too when dieing, so that maybe isn't a huge deal either way.
So I'd say get DST when you need to choose. Imho it's the superior game when we're talking raw (DLC-/mod-less) base content but also usability and QoL. If you're happy with DST and wish to see the big and small differences yourself (or if you want to play Shipwrecked and Hamlet without using mods) then you could still always get DS further down the line.
If you want more and better content, get singleplayer.
Both go on sale frequently too, so you can always get both.
