Basically you almost always need to pick a non-starter weapon first to be able to discover secret rooms reliably, and then you pick another item for a total of 2 non-starters, at which point the game starts getting harder and the lack of items really starts to hit you. I can't do Mines+ with just three items, no health upgrades, no nothing.
As for finding secret rooms, they're much less important come rainbow mode as you can't even open chests inside them.
Maybe it's just my experience, I just generally find rainbow runs far easier overall and usually end up with some crazy builds if I choose the right items.

The A-S Tier modifier you're talking about only affects regular, randomly encountered rainbow chests. Either way, I managed to beat the mode by simply brute-forcing my way through it in a regular Pilot run. Thanks for the help. Cheers!
