Edit: there is no new game plus and some of the collectibles you may collect after some story progression
I don't remember how the saves work so play like you have to do everything in one run.
Also some quest might seem like you can do them infinite amount of times but they are not infinite ( around 20 - 30 boring ones from one person that gives you quest I think).
My idea is to play for like 2h and see what you need to do for 100%.
If you have more questions just ask.
Do the side missions first when you can, you get some good bumps from the girlfriend missions in particular. Also other side missions like retrieving the green idols unlock more moves in the martial arts studio.
Each story mission gives you a rating that adds to the blue and red meters, if you really screw one up you might consider reloading a save and playing through the mission again to get a bigger bump.
The definitive edition doesn't have DLCs.
It does. You start them from main menu.
