Are you talking about old school Doom where you could setup a multiplayer game on your local LAN?
Then no, sorry.
Steam BTD5 Co-op is only an online, 2 player game. Period. Online access is mandatory for Co-op play.
You can match up with people randomly or you can also create Private Co-op games with people who are on your Steam Friends list.
Steam BTD5 solo play does not require an active connection, though to save in the cloud and such things you obviously do need to be online.
